Biochemical and molecular identification of probiotic bacteria isolated from traditional dairy products of Eastern Azerbaijan

Probitics are dietary supplement of live microorganisms which confer a health benefit on the host when administrated in adequate amount in gastrointestinal system. Lactic acid bacteria (LAB) are the most common type of microbes used as probiotic. Traditional dairy products are the main source of probiotic bacteria. Isolation and identification of probiotic bacteria from these products are of high importance. At the present study identification of 97 isolates was performed, using biochemicall and molecularly strategies. According to the biochemical assays such as cell morphology, Gram staining, sugar fermentation pattern, ability to grow at different range of pH, tempreture and NaCl, and grouped by Ntsys software. Based on biochemical similarity, Lactobacillus isolates were grouped as: A: casei-maltaromicus- sakei, B: buchnerii- brevis- reouteri- fermentum, C: acidophilus- delbruchi- brevis, D: plantarum- agilis- morinuri and Enterococcus isolates were divided to a: fesium, b: fecalis and c: durans-hirae. For further investigations, the isolates were studied based on PCR-RAPD –ARDRA, Sequencing technics. Out of group A, one isolate was selected and sequenced. It was attributed to casei species. Out of group B three isolates were considered as indice of which two isolates were recongnised as brevis specises by sequencing and the other as Lactobacillus sp. out of group C, three isolates were chosen and determined as brevis species. Moreover, out of group D, five isolates were sequenced and identified as plantarum species. From Enterococcus isolates, two isolates of group a, three isolates of group b and two isolates of group c were selected and sequenced. According to molecular data the biochemical results were confirmed in two isolates of faecalis and fecium species.  However one of the selected isolte from group c was introduced as durans species and one other isolate showed high mutation in 16S rDNA gene. These isolates were repectively submitted as Enterococcus sp.
